Abstract | SMT1-1 is a reference material (RM) designed for the development of SARS-CoV-2 spike glycoprotein detection methods, as well as an antigen source for use in SARS-CoV-2 immunological assays. The majority of available SARS-CoV-2 test kits and vaccines exploit the interaction between the SARS-CoV-2 spike glycoprotein and human anti-spike antibodies. Reference values for the molar concentration, mass fraction, and mass concentration of SARS-CoV-2 spike glycoprotein in SMT1-1 have been established. Values are provided for the known spike protein sequence region that ignores the glycan, and also the full spike glycoprotein where the mass fraction and mass concentration values include an estimate of the molar mass of the glycan. Information values for the size heterogeneity of SARS-CoV-2 spike glycoprotein in SMT1-1 are provided. A unit of SMT1-1 contains 1.2 mL of SARS-CoV-2 spike glycoprotein in Dulbecco's phosphate-buffered saline (DPBS) with 10 mM HEPES sodium salt. |
